چکیده

is essentially irreversible. In free solution the reaction is slow because of spin inversion associated with the reaction of singlet reduced flavin with triplet 0, and complex, as described below. With flavoproteins the reaction with 0, may be orders of magnitude faster or slower, depending on the specific flavoprotein or the class of flavoprotein to which i t belongs. Clearly the reactivity of the reduced flavin with 0, is modulated by the protein environment in which i t is located, both in terms of rates and products of the reaction (see Ref. 1 for a more detailed review).

New flavins for old: artificial flavins as active site probes of flavoproteins.

Flavin enzymes are unique as biological catalysts in that they carry out a wide variety of different biochemical processes. This sets them apart from enzymes which use other coenzymes, like pyridine nucleotides, pyridoxal phosphate or thiamin pyrophosphate, each of which involves a single type of chemical event. Flavins thus are involved in the dehydrogenation of a variety of substrates, in one...

Mechanisms of Reaction of Some Flavoprotein Enzymes with Oxygen

Biochemical investigations of the properties of free flavins and of flavoproteins have shown that reduction usually occurs in two stages, with the intermediate formation of semiquinones in the case of free flavins. Flavoproteins often show spectroscopically similar intermediates, when partially reduced with substrate. These may, however, be enzyme-product complexes. Detailed investigation of in...

The chemical and biological versatility of riboflavin.

Since their discovery and chemical characterization in the 1930s, flavins have been recognized as being capable of both one- and two-electron transfer processes, and as playing a pivotal role in coupling the two-electron oxidation of most organic substrates to the one-electron transfers of the respiratory chain. In addition, they are now known as versatile compounds that can function as electro...

EPR on Flavoproteins.

Flavoproteins often employ radical mechanisms in their enzymatic reactions. This involves paramagnetic species, which can ideally be investigated with electron paramagnetic resonance (EPR) spectroscopy. In this chapter we focus on the example of flavin-based photoreceptors and discuss, how different EPR methods have been used to extract information about the flavin radical's electronic state, i...

Photoexcited structure of a plant photoreceptor domain reveals a light-driven molecular switch.

The phototropins are flavoprotein kinases that control phototropic bending, light-induced chloroplast movement, and stomatal opening in plants. Two flavin mononucleotide binding light, oxygen, or voltage (LOV) domains are the sites for initial photochemistry in these blue light photoreceptors. We have determined the steady state, photoexcited crystal structure of a flavin-bound LOV domain. The ...
